Are you on manual or auto sync? You might have files in your library that cannot be played on the iPod (streaming songs files are one example) or you have songs from the iTunes Music Store that haven't been authorized for play yet so they weren't transferred. You can view file Kind by right clicking on the info bar at the top of iTunes. If you have any Music Store files, just double click one to play and enter your ID and password when asked.
